The role of the light source in an atomic absorption spectrophotometer is to provide characteristic spectral lines (resonance lines) of the element to be measured. The light source is required to emit sharp resonance lines, have sufficient radiation intensity, low background, good stability, low noise, easy operation, and long service life. The commonly used sharp line light source is a hollow cathode lamp, which is a special gas discharge tube mainly composed of a tungsten rod anode and a hollow cathode made of pure metal of the measured element.
Characteristics of atomic absorption spectrophotometer:
1. High sensitivity: The relative sensitivity of atomic absorption spectrophotometry for determining most metal elements is 1.0 × 10-8-1.0 × 10-10g • mL-1, while the sensitivity of non flame atomic absorption spectrophotometry is 1.0 × 10-12-1.0 × 10-14g. This is because atomic absorption spectrophotometry measures the ground state atoms, which account for more than 99% of the total number of atoms, while atomic emission spectroscopy measures the excited state atoms, which account for less than 1% of the total number of atoms. Therefore, the sensitivity and accuracy of the former are much higher than those of the latter.
2. Good precision: Due to the small impact of temperature changes on the measurement, this method has good stability and reproducibility, with good precision. The relative standard deviation of general instruments is 1% -3%, while instruments with good performance can reach 0.1% -0.5%.
3. Strong selectivity: Due to the fact that atomic absorption spectral lines only occur in the main line system and are very narrow, the probability of line overlap is much smaller than that of emission spectra, resulting in less spectral interference and stronger selectivity. Moreover, spectral interference is easy to overcome. In most cases, coexisting elements do not interfere with atomic absorption spectra. Due to its strong selectivity, the analysis is accurate and fast. Analyzing an element only takes tens of seconds to minutes.
Precautions for atomic absorption spectrophotometer:
1. The instrument should be placed in a dry room and used on a sturdy and stable workbench. Indoor lighting should not be too strong. During hot weather, it is not advisable to use an electric fan to blow air directly onto the instrument to prevent unstable illumination of the bulb filament.
Before using this instrument, the user should first understand the structure and working principle of the instrument, as well as the functions of each control knob. Before turning on the power, the safety performance of the instrument should be checked. The power wiring should be firm, and the power supply should be good. The starting position of each adjustment knob should be correct before turning on the power switch.
When the instrument is not yet powered on, the pointer of the electric meter must be on the "0" mark. If this is not the case, the calibration screw on the electric meter can be used to adjust it.
