Tuner (DE3) Escherichia coli host strain: This strain is derived from BL21 (DE3) strain and was obtained by mutating the lacZY gene (galactosidase gene) on the genome of BL21 (DE3) strain. This mutation allows IPTG from the external environment to enter each E. coli cell in the culture medium at a uniform rate, resulting in a more stringent and uniform concentration dependence of cells on IPTG.

This strain is derived from the BL21 (DE3) strain and was obtained by mutating the lacZY gene (galactosidase gene) on the genome of the BL21 (DE3) strain. This mutation allows IPTG from the external environment to enter each E. coli cell in the culture medium at a uniform rate, resulting in a more stringent and uniform concentration dependence of cells on IPTG.

Like its parent strain, this strain integrates a DE3 region on its chromosome, encoding T7 bacteriophage RNA polymerase. When used in conjunction with a vector containing the T7 promoter, the expression level of exogenous proteins on the vector can be adjusted from low to ultra-high levels by precisely adjusting the concentration of IPTG. Especially at low levels of expression, protein solubility may be better.
