Interleukin-15 (IL-15)It is a T cell growth factor that was discovered by two independent laboratories in 1994 and has similar biological functions to IL-2. IL-15 is mainly produced by monocytes and macrophages. IL-15 mRNA is expressed in various tissues of the human body, such as the heart, lungs, kidneys, especially the placenta and muscles. The cell source and target cell distribution are wider than IL-2, so it may exert a similar effect to IL-2 in areas where IL-2 is not expressed.

The mechanism of action of IL-15:
IL-15R α is widely expressed in humans and mice. The main mechanism of the IL-15 signaling pathway is that IL-15 binds to the IL-15R α subunit on cells, and IL-15 stays on the cell surface, forming immune synapses with IL-2R/IL-15R β - γ c on nearby effector NK cells or T cells, activating the JAK1/JAK3 and STAT3/STAT5 pathways, Syk kinase and phospholipase C (PLC) γ, Lck kinase and Shc, leading to the activation of PI3K/Akt and Ras/Raf/MAPK signaling cascades. These pathways lead to subsequent expression of Bcl2, Myc, Fos/Jun, and NFkB activation.

Biological functions and applications of IL-15:
IL-15 and IL-2 have similar biological functions. IL-15 and IL-2 share the gamma c-receptor subunit, and both cytokines can induce the differentiation of T cells, B cells, and natural killer cells (NK cells) and promote their proliferation. At present, IL-15 is often used in combination with IL-2 for in vitro expansion and culture of T cells or NK cells.
It is worth noting that IL-2 can cause excessive differentiation of T cells, forming aging T cells with weaker killing ability, and can induce apoptosis of activated T cells. The advantage of IL-15 in immune cell therapy is that it does not cause activated T cell apoptosis.
