The basis of antibody affinity purification is the high affinity and specificity of protein G and protein A for the Fc region of IgG from different species. Fixing protein G or protein A on chromatography media of different matrices provides a method for rapidly separating IgG and IgG subclasses from ascites, cell culture supernatant, and serum.

At the same time, we also have chromatography media for recombinant protein L ligands for the purification of antibodies and antibody fragments. Protein L has a strong affinity for the variable region of kappa light chains and can be widely used to capture antibody fragments such as Fab fragments, single chain variable region fragments (scFv), and binding domain antibodies (Dabs).

GE affinity chromatography packing is a technique for separation based on reversible interactions between target proteins and specific ligands on the guiding matrix. This interaction can be either specific (such as the binding of antibodies to protein A or protein G) or non-specific (such as the binding of tagged proteins to metal ions).
There are many types of biological interactions applied in affinity chromatography, such as ligand substrate, protein protein, nucleic acid protein, and so on. The affinity chromatography process generally includes four stages: equilibrium, sample loading, cleaning, and elution. Balance mainly refers to balancing the column in an environment that is conducive to the binding of the sample and filler, and then loading the sample. Elution can be specific (such as competitive ligand molecules) or non-specific (such as altering pH or ionic strength). Affinity chromatography has the advantage of high selectivity, and usually a single affinity chromatography column can obtain high-purity products. However, when higher purity is required, one or more purification steps need to be added, such as purification after gel filtration chromatography.
Affinity chromatography packing can be used for purifying tag proteins, antibodies, or specific molecules. Affinity chromatography can also be used to remove specific pollutants.
General Electric Medical Group recently launched a new affinity chromatography medium, Capto L, suitable for capturing antibodies and antibody fragments. It combines a rigid high flow agarose matrix with a recombinant protein L ligand that binds to immunoglobulin, which has strong affinity for the variable region on the antibody K light chain. Therefore, Capto L is suitable for capturing various antibody fragments, such as Fab, single chain antibodies (scFv), and domain antibodies.
Compared with ion exchange and hydrophobic interaction chromatography, the affinity chromatography capture step has a very stable and universal processing procedure. The optimization limit required for Capto L purification is small, and a high purity can be obtained in one step. This significantly saves costs, time, and efficiency during the production process. Compared with other methods, the high selectivity of the affinity chromatography capture step will help reduce the subsequent fine purification steps, and fewer selective purification media will be used in production, which can also benefit from it.
