SpectraMax Gemini ® EM is a full wavelength fluorescence enzyme-linked immunosorbent assay (ELISA) developed by Molecular Devices. Through its spectral scanning function, it can determine the optimal excitation and emission wavelengths for unknown fluorescent dye molecules, SpectraMax Gemini ® EM uses a dual grating monochromator to detect various novel fluorescent dye molecules without the need to purchase expensive filters. Whether it is for detecting uniform solution fluorescence signals or determining the fluorescence intensity of adherent cells, SpectraMax Gemini ® The top and bottom dual fluorescence light paths of EM can be easily handled and freely switched. SpectraMax Gemini ® EM has a single well multi-point scanning mode, which can read multiple points per well. When applied to cell experiments, it can achieve higher levels of sensitivity. Auto PMT (photomultiplier tube automatic gain adjustment technology) can effectively expand the range of detection dynamics, avoiding the occurrence of photobleaching and fluorescence supersaturation phenomena. In addition, the instrument is equipped with two secondary detection modes, chemiluminescence and time-resolved fluorescence, which facilitate users to expand to more detection methods under limited resources.

Experimental examples
We know that more sensitive methods are needed for next-generation sequencing and amplification product quantification. Life Technologies' Quant iT PicoGreen dsDNA experiment has better specificity for DNA and is 1000 times more sensitive than traditional absorption light methods. To maximize the sensitivity of the experiment, it is necessary to use the optimal excitation and emission wavelengths. SpectraMax Gemini ® The EM type multifunctional microplate reader does not have the dual grating of color filter based readers, which allows users to select any wavelength within the nominal range of the instrument. It is important to determine the excitation and emission wavelengths that can generate the optimal experimental dynamic range. The sensitivity observed using the 96 well microplate format is as low as 63 pg/mL, which is significantly lower than the nominal 250 pg/mL in the QuantiT PicoGreen experimental product manual.
