Preparation of mobile phase:

A liquid chromatography column is used for separating sample components through mass exchange between the column packing and the mobile phase. Therefore, the mobile phase is required to have the following characteristics:

a、 The flowing relative sample has a certain solubility, ensuring that the sample components do not precipitate in the column (or remain in the column for a long time).

b、 The mobile phase has a certain degree of inertness and does not undergo chemical reactions with the sample (except in special circumstances).

c、 The viscosity of the mobile phase should be kept as low as possible in order to achieve good separation results when using longer analytical columns; Simultaneously reducing column pressure drop and extending the service life of the liquid pump (the method of increasing temperature can be used to reduce the viscosity of the mobile phase).

d、 The physicochemical properties of the mobile phase should be adapted to the detector used. If using a UV detector, ZUIHAO is prepared with a solvent that has low UV absorption.

e、 The boiling point of the mobile phase should not be too low, otherwise bubbles may be generated, making the experiment impossible to proceed.

f、 After preparing the mobile phase, it is necessary to perform degassing. Removing trace gases dissolved in the mobile phase is beneficial for detection and can also prevent trace oxygen in the mobile phase from reacting with the sample.

Liquid chromatography columns have also been commercialized. Due to its unique properties, it is generally used for special purposes. For example, graphitized carbon is gradually becoming a packing material for reverse phase chromatography columns. The separation of this filler is different from that of silicone based alkyl bonded phases, where the surface of graphitized carbon is the retained basis and no further surface modification is required. The retention capacity of the column packing is generally stronger than that of alkyl bonded silica gel or porous polymer packing. Graphitic carbon can be used to separate certain geometric isomers, and due to its insolubility in HPLC mobile phase, this type of column can be used at any pH and temperature.
